N4-acetylcytidine (ac4C) is a highly conserved RNA acetylation modification that is widely present in tRNA, rRNA, and mRNA of both eukaryotes and prokaryotes. It is significantly associated with a variety of human diseases, especially cancer. RNA acetylation may play a crucial role in regulating the initiation and elongation of protein translation, enhancing the accuracy of protein translation, and improving mRNA stability and translation efficiency.
Notably, the ac4C modification does not affect complementary CG base pairs and cannot be detected by conventional RNA sequencing. Acetylated RNA immunoprecipitation (acRIP) enriches ac4C-containing RNA using a specific antibody. Subsequently, by combining with qPCR (acRIP-PCR) or high-throughput sequencing (acRIP-seq), it enables accurate analysis of the information regarding RNA ac4C modification sites.
